I'd love it if the opportunity to play as Joker became available. It'd be fun if when selecting a choice - Joker does the exact opposite. It'd show the unpredictability of the personality as well as gain some insight into what could him tick. But that's just me.
Also off topic - Will the game be a 5 story arc or stand alone episodes? I'd prefer the latter.
An interesting character to play as would be Amanda Waller. It'd be like a management sim in which you have to make lots of important decisions to guide the Suicide Squad to victory. You'd have to decide what risks to take, what leads to pursue and which of your team-mates to sacrifice at crucial moments. You'd also have to decide if you're going to be a ruthless (shoot all my own damn employees for knowing too much) Waller, or a more "compassionate" Waller who values innocent life.
Waller would spend most of the game in her control centre, but she'd see the action unfold through the eyes of the Suicide Squad members, who would be equipped with body-cams, and a gameplay mechanic might even be to be able to jump between these cams at will.
An example of a big decision/order you might be forced to give is:
Killer Croc vanished into the sewers in the heat of battle, and he's not responding to hails.
Option Y: Do nothing. See how it pans out.
Option X: Order the entire Suicide Squad to pursue and recover him, even though time running out.
Option A: Order Deadshot to break away and pursue Croc alone.
Option B: Detonate Croc's suicide collar/neck bomb.

I really want to play as Catwoman, or at least see her in a leading role of her own after Batman.
Telltale has made Selina such a dynamic, nuanced, and enjoyable character that I think it would be really awesome to see her lead a mini-series like Michonne's.
And Laura Bailey deserves more opportunities to play Catwoman. Her performance is simply phenomenal.
