Lee said he hadn't seen his family in a long time. Plus, when you looked at the pictures displayed in the drugstore, there were some with Lee's parents and brother, but I'm pretty certain none of them with Lee. So I believe he didn't feel like he was part of the family anymore.
The first is that he didn't want the rest of the group to figure out who he was, and that he was a convicted murderer. They could potentially cast him out of the group over it, which could make his (and Clem's) situation a lot more precarious.
The second is that he was ashamed of what he had done and the hurt it had causes his family. The murder seemed to have caused a rift between Lee and his family, as Lee can mention to Clem that they hadn't spoken in awhile. He also clearly regrets this. Tearing himself out of the family photo was as much an expression of shame and anger over what he had done, as it was an act of self preservation.
After you see Lee's brother trapped on the other side of the street, Doug wants you to get proof that he works in the pharmacy.. Obviously Lee can't tell him that it's his brother, so when he gets back with the family photo, he wont notice Lee in it.
