I did that to Lee and still...
He turned to a walker (well, almost), though, Clementine put Lee to rest because it was the right thing to do.
Supposedly chopping off of the arm should also remove the infection, but it spread by that time which made it pointless to saw his arm off.
Episode 5 kind of felt pretty fast compared to the others which lasted longer, but I'll be sure to replay it again.
Note: I thought chopping off the arm would solve Lee's problem because the TV series proved that the infection would stop if the arm/leg were taken apart.

It probably took over an hour to tread to the shelter... (speculation, but I doubt it was a fast journey) - By then it was too late. Way too late.
By the time the option to amputate comes up, I figured it was already too late. In the cases where amputation worked, it's because it was done immediately after the bite. The fact Lee passed out kinda told me the infection had spread which meant that amputation was pointless in that case.
They just kinda dithered too long; one of my minor gripes actually, Lee wasn't exactly moving with a purpose. "I'll probably be dead in a few hours, but I don't need to run or even walk like I need to get crap done".
Normally I'd say to avoid the tv show for logic on the game(as it's based off of the comics), but they both seem to have treated amputation the same. Anyway, notice that Hershal(or if we are going with the comics, Dale) got amputated immediately. Rick didn't wait a half hour like Lee did, that's why it failed.
*COMIC BOOK SPOILERS*
Actually, it took some time until Dale had his leg cut off, he was bit in the parking lot, he had to crawl out of there and catch up to Herschell's son, then go to the infirmary and only THEN have his leg amputated. The two characters who had their limbs amputated instantly (Allen and Morgan) died, because they lost too much blood before receiving medical attention.
So I don‘t think amputation has to be done two seconds after you were bit or you‘re done. It‘s also uncertain if Lee died because of the bite, or from blood loss or an infection in his putated arm.
Lee likely died from the bite, as he feels the same sensations from it both before and after losing his arm.
It's pretty clear it was the bite that did him in, Lee passed out before the option to amputate comes up. That's a pretty big clue that it spread and that by that point amputation was basically just rearranging chairs on the Titanic.
I think we can safely rule out another infection after amputation - it wouldn't have killed him that quickly. Nor is Amputated Lee leaving a red trail wherever he goes, he's not sitting in a pool of his own blood at the end, etc. aside from the arm, he's identical to the two-armed Lee thoughout the episode and dies as the exact same time. Only logical conclusion is that they died of the same thing.
I still cut off his arm in my playthrough, I still had hope XD
